摘要
为进一步提高菌株产中性蛋白酶能力,在原生质体最佳形成条件下制备得到Bacillus subtilis UN9原生质体,并对其进行紫外线与亚硝基胍复合诱变,通过摇瓶复筛,得到高产、稳定的突变株Bacillus subtilis Promax NTG14,产酶能力相比出发菌株提高了近24%,中性蛋白酶活力达4286.09U/mL。试验结果表明,复合诱变原生质体是一种快速、有效的优良产酶菌株选育途径。
In order to improve the neutral proteinase (NPR) productivity of Bacillus subtilis UN9, protoplasm from B. subtilis UN9 were prepared under its optimal formation conditions and mutated multiply by ultraviolet radiation and nitrosoguanidine (NTG). A stable, highly-yield NPR strain named B. subtilis Promax NTG14 was gained with an improvement of 24% relative to primordial strain, and its enzyme activity Can reach 4286.09 U/ml. Experimental results indicated that the combinated mutation of protoplast is a quick and efficient approach for screening the excellent NPR-producing strain.
