摘要
目的:探讨华蟾素对人胆管癌细胞株QBC939的抑制作用及可能的作用机制。方法:流式细胞仪检测华蟾素对QBC939细胞凋亡的作用,RT-PCR法研究华蟾素对人胆管癌细胞株QBC939 Fas及caspase-3 mRNA表达变化的影响。结果:不同浓度华蟾素作用于QBC939细胞48 h后,流式细胞仪分析显示,凋亡细胞明显增多,与对照组相比有显著性(P<0.001)。RT-PCR结果显示:Fas、caspase-3 mRNA表达实验组与对照组相比增强(P<0.001)。结论:华蟾素作用于人胆管癌细胞株QBC939,促进其细胞凋亡,而这一作用的实现可能与Fas及caspase-3 mRNA表达的上调有关。
Objective To investigate the gTowth inhibition effect of Cinobufacini on cholangiocarcinoma cell QBC939 in vitro and possible mechanism. Methods Cholangiocarcinoma cell line QBC939 were cultured in vitro. Apoptosis in the cholangiocarcinoma cell QBC939 after different doses of cinobufascini administration were evaluated by flow cytometry.The effect of cinobufacini on the expression of caspase - 3 and Fas mRNA were evaluated by RT - PCR. Results The result of the flow cytometry showed that cinobufacini could induce cell apoptosis. Expression of Fas, caspase - 3 mRNA depended on the dose of cinobufacini. Cinobufacini could increasi its expression. Conclusion Cinobufacini induces apoptosis of human extrahepatic cholangiocarcinoma cell line QBC939 probably through the mechanism of the up - regulation of expression of Fas and caspase- 3 mRNA.
