摘要
研究了CTAB/正戊醇-正辛烷反胶团溶液萃取分离螺旋藻藻蓝蛋白的性能.考察了水相离子强度和种类,有机相中表面活性剂浓度和助溶剂浓度等因素对萃取行为的影响,并从反胶团的微观结构予以解释.结果表明:0.04 mol/L CTAB/正戊醇-正辛烷(体积比1∶4)的反胶团体系用于萃取pH7.0,包含0.1 mol/L KCl的螺旋藻细胞破碎液,藻蓝蛋白萃取率可达96.3%,分配系数达到26.0;采用pH5.0,包含2 mol/L KBr的反萃液反萃藻蓝蛋白,反萃取率可达90.6%.
The supematant extraction of c-phycocyanin by reversed micelles using surfactant CTAB was carried out. Various parameters of the aqueous phase (pH, the type and concentration of salt ), as well as the organic phase (concentration of surfactant and co-solvent )were studied. The experimental data can be interpreted reasonably by the microscopic structure of the micellar organic phase. It is reported that from the supematant of pH7.0,0.1 mol/L KC1 disrupte Spirulina cell, the partition coefficient of c-phycocynain is as high as 26.0 with a yield of 96.3% extracted by 0.04 mol/L CTAB/n-pentanol-octane ( 1 : 4v/v ) reversed micelles. A back extraction yield of c-phycocynain is 90.6% by pHS.0, 2 mol/L KBr solution as back extractant.
