摘要 学术背景:干细胞是一群较原始的细胞,它们具有自我更新和多向分化潜能,可以被诱导分化为胰岛素分泌细胞,理论上可提供丰富的胰岛细胞来源。目的:本文就近年来干细胞诱导分化为胰岛素分泌细胞的研究进展做一综述。检索策略:应用计算机检索Pubmed和CNKI数据库1990-2007年期间相关文献,检索词为"干细胞,诱导分化,胰岛素分泌细胞,stem cell,differentiation,islet-producing cell"。对资料进行初审,并查看每篇文献后的引文。纳入标准:干细胞向胰岛素分泌细胞诱导分化的研究进展。排除标准:重复研究。文献评价:共收集到相关文献162篇,选择其中49篇文献进行重点阅读和分析。49篇文献中,1篇涉及糖尿病及其并发症的治疗,1篇涉及胰岛细胞移植,1篇涉及干细胞在糖尿病治疗中的应用,13篇涉及胚胎干细胞诱导分化为胰岛素分泌细胞的研究,4篇涉及脐血来源的干细胞体内分化为胰岛素分泌细胞的研究,10篇涉及骨髓来源的干细胞分化为胰岛素分泌细胞的研究,13篇涉及胰腺干细胞分化为胰岛素分泌细胞的研究,6篇涉及其它器官来源的干细胞分化为胰岛素分泌细胞的研究。资料综合:胰岛细胞移植可以用来治疗1型和部分2型糖尿病,但由于供体细胞来源匮乏,因而寻找新的细胞来源成了当务之急。近年来,干细胞研究为新型胰岛细胞的来源带来了希望。目前用于胰岛细胞来源研究的干细胞主要有胚胎干细胞和成体干细胞,后者根据细胞来源不同分为脐血来源干细胞、骨髓来源干细胞、胰腺干细胞及其他器官来源的干细胞。目前的研究显示,骨髓来源的干细胞可以在体内促进胰腺的再生,并且可以在体外被诱导分化为胰岛样细胞。结论:深入了解干细胞向胰岛β细胞诱导分化的分子调控机制,将会加快糖尿病细胞治疗的研究进展。 BACKGROUND: Stem cells are a flock of initial cells, which have capabilities of self-renewal and multidifferentiation, and can be differentiated into islet-producing cells. Theoretically, stem cells are a potential affluent source for islet cells. OBJECTIVE: This article is a review about the research progress of the differentiation from stem cells into islet-producing cells. RETRIEVAL STRATEGY: Pubmed and CNKI databases were undertaken to identify the relevant articles published from 1990 to 2007 with the key words of "stem cell, differentiation, islet-producing cell" in English and in Chinese. The data were selected firstly, and each citation was checked in each article. Inclusive criterion: Articles about research progress of the differentiation from stem cells into islet-producing cells were selected. Exclusive criterion: The repetitive studies were excluded. LITERATURE EVALUATION: Totally 162 relevant articles were collected and 49 papers of them were carefully read and analyzed. Of the 49 inclusive articles, 1 was about the treatment of the diabetes and the complications, 1 about islet transplantation, 1 about application of the stem cells in the treatment of the diabetes, 13 about the research on the differentiation of embryonic stem cells into islet-producing cells, 4 about the research on differentiation of umbilical cord blood derived stem cells into islet-producing cells, 10 about the research on the differentiation of bone marrow derived stem cells into islet-producing cells, 13 about the research on differentiation of pancreatic stem cells into islet-producing cells, and 6 about the research on the differentiation of other organ-bound stem cells into islet-producing cells. DATA SYNTIIESIS: The transplantation of pancreatic islet cells has been used to treat the diabetes type l and 2.
