摘要
将供试9个青枯菌菌株在TTC选择性培养基平板上纯化,移植于扩繁培养基上培养,根据Hay-ward划分青枯菌生化型的标准,将其分别接种在含糖、含醇化合物的鉴定培养基上,观察其对3种双糖和3种己醇的氧化利用能力从而对其归类划型.由于不同生化型的青枯菌对3种糖、3种醇化合物的利用情况不同,因而可根据培养基颜色(pH)发生的变化来确定供试菌株属于哪一个生化型.结果表明:9个供试青枯菌菌株分别属于5个生化型,即生化型Ⅰ、Ⅱ、Ⅲ、Ⅳ、Ⅴ.其中大部分属于生化型Ⅰ,它们是DA1、DB1、DC1、DD1,占44.44%;UW 551和P041属于生化型Ⅱ,占22.22%;T032属于生化型Ⅲ,ZO4属于生化型Ⅳ,M5属于生化型Ⅴ,各占11.11%.
Nine strains of Ralstonia Solanacearnm were purificated on the TTC alternative culture medium and then transplanted to multiplicate in liquid medium. With sterile water we diluted the concentration of bacteria suspension to 3 ×10^8cfu/ml. These hio- type strains were used for classification based on Hayward's classification criterion of P. solanacearnm. They were examed oxidative utilization of three kinds of disaccharide ( lactose, maltose, cellobio) and three kinds of hexylalcohal ( mannitol, sorhitol, dulcitol). As different types of biochemical variant have different results on three kinds of sugar and three kinds of alcohol. Therefore, the col- or of the culture (pH) have different changes. We can determine which biochemical variant it belongs to. The results showed that nine Ralstonia Solanacearnm tested separately belonged to five hiovars. They were hiovar Ⅰ、Ⅱ、Ⅲ、Ⅳ、Ⅴ. The main biochemical strains of R. Solanacearum among them belonged to hiotype Ⅰ (44.44%) , and they were DA1 ,DB1 ,DC1 and DD1. UW551 and PIM1 belonged to biotype Ⅱ (22.22%). T032,ZO4 and M5 seperately belonged to biotype Ⅲ, biotype IVand biotype Ⅴ. The percentages of them were all 11.11%.
