摘要
目的:检测食管鳞状细胞癌(ESCC)组织中错配修复基因hMLH1、hMSH2及P53的表达。方法:采用免疫组织化学SP法检测62例ESCC组织、31例癌旁不典型增生组织及62例正常食管黏膜组织中hMLH1、hMSH2及P53蛋白的表达。结果:ESCC组织、癌旁不典型增生组织与正常食管黏膜组织中hMLH1的阳性表达率分别为27.4%,58.1%及88.7%,hMSH2的阳性表达率分别为32.3%,51.6%及91.9%,P53的阳性表达率分别为79.0%,54.8%及35.5%,3种组织中hMLH1、hMSH2及P53的阳性表达率差异均有统计学意义(P<0.05)。ESCC组织中hMLH1、hMSH2及P53的表达与分化程度、浸润深度及有无淋巴结转移有关(P均<0.05);hMLH1、hMSH2阳性表达者P53的阳性率均低于hMLH1、hMSH2阴性表达者。结论:ESCC组织中存在hMLH1与hMSH2的低表达及P53的高表达,3者可能在食管癌的浸润、转移及黏膜上皮癌变过程中起重要作用。
Aim : To investigate the expression of human mismatch repair gene(hMLH1/hMSH2) and P53 in esophageal squamous cell carcinoma (ESCC) tissue and its significance. Methods:SP immunohistochemieal method was used to detect the expression of hMLH1 ,hMSH2 and P53 protein in 62 cases of ESCC specimens, 31 cases of adjacent atypical hyper- plasia epithelium and 62 cases of normal esophageal epithelium. Results: The positive expression rates of hMLH1 in ES- CC, adjacent atypical hyperplasia epithelium and normal esophageal epithelium were 27.4% , 58. 1% and 88.7% , and those of hMSH2 were 32.3% , 51.6 % and 91.9% , and those of P53 were 79.0% ,54.8% and 35.5% , respectively. The expression of hMLH1, hMSH2 and P53 protein was associated with the differentiation levels, depth of invasion and lymph node metastasis of ESCC (P 〈 0.05). The expression rate of P53 in cases with negative expression of hMLH1 and hMSH2 was higher than those with positive expression of hMLH1 and hMSH2. Conclusion : hMLH1, hMSH2 and P53 play important roles in the infiltration, metastasis and carcinomatous change of the mucosa epithelium in ESCC.
