摘要
目的:观察肺纤方(Feixian Formula)对博来霉素致肺间质纤维化大鼠生存状态的影响。方法:85只雄性Wistar大鼠经气管插管灌注博莱霉素(1mg/kg体质量)复制大鼠肺间质纤维化模型,随机分入模型组29只、泼尼松组28只、肺纤方组28只;15只雄性Wistar大鼠经气管插管灌注等体积生理盐水作为假手术组。造模后24时开始常规每日一次灌胃给药,泼尼松组给予醋酸泼尼松5mg/kg体质量,肺纤方组给予肺纤方颗粒1.25g/kg体质量(相当于生药4.475g/kg体质量),模型组和假手术组给予等体积生理盐水。实验全程观察大鼠生活状态如活泼程度、毛发、饮水量、进食量的变化,有无出血、腹泻、死亡,有无其它异常情况;同时监测大鼠体质量变化。结果:实验全过程中,肺纤方组和假手术组没有死亡大鼠,模型组死亡4只大鼠,泼尼松组死亡2只,P=0.1304;大鼠体质量变化显示模型组体质量增长更快,从第21天开始,模型组大鼠体质量与假手术组比较没有统计学意义,P>0.05;综合评价认为肺纤方组大鼠的整体状态好于模型组和泼尼松组。结论:肺纤方能够改善大鼠的整体状态。
Objective: To study the influence of Feixian Formula on general state of bleomycin-induced pulmonary fibrosis model rats.
Method: Totally 85 male Wistar rats were infused with bleomycin (1 mg/kg body weight) through tracheal intubation to form pulmonary fibrosis, and they were randomly and equally divided into model group 29, prednisone group 28, and Feixian Formula group 28. Other 15 male Wistar rats were infused with equivalent saline through tracheal intubation as sham opetation group. Twenty-four hours after operation, intragastric administration of prednisone (5mg/kg) and Feixian Formula (1.25g/kg, i.e. raw material 4.475g/kg) was given to prednisone group and Feixian Formula group respectively once a day. Equivalent saline was administrated to model and sham operation groups. In the whole experiment, the general state of rats were observed such as spirit, appetite, activity, fur, weight, death, and any clinical situation that is possible. And weight changes were observed. On the 14th , 28th and 45th day.
Results: In the 45 days experiment there was no death case in FXP group and fake operation group, 4 death case in model group, and 2 in prednisone control group, however, the statistics shows P=0.1304. The weight changes showed that the weight in model group increase more quickly, for from the 21st day there was no significant between model group and sham operation group, P〉0.05. The general state of Feixian Formula group was better than that of model and prednisone group.
Conclusion: Feixian Formula could improve the general state of rats that were infused with bleomycin to cause pulmonary fibrosis.
