摘要
目的:研究单次持续40%幅度的牵拉作用于人皮肤成纤维细胞后bFGF、TGFβ1的表达,初步探讨机械应力作用下细胞增殖的机理。方法:体外培养人正常皮肤成纤维细胞,将细胞转移至牵拉细胞培养模型中的弹性膜上,待细胞贴壁生长至75%~80%融合时,实施40%单次持续牵拉,用WESTERN-BLOT对非牵拉组与牵拉组24h、36h、48h、72h的细胞bFGF、TGFβ1进行检测。结果:牵拉各组及对照组均呈阳性表达,但牵拉48h后两者的表达较未牵拉时明显增加。结论:单次40%持续的牵拉方式可以促进bFGF、TGFβ1表达,可能是导致细胞的增殖。
Objective To study the expression of bFGF,TGFβ1 of human skin fibroblasts treated by mechanical stress. Methods A new cell culture model was designed, which can load mechanical strain. Primary cultured skin fibroblasts were obtained from full thickness skin residual in skin-graft surgery. The fibroblasts were planted on elastic membranes of the model. After the 75%-80% confluence of the cell was reached, mechanical stretch was applied. The extend degree was 40% in the experiment and after stretching for 24h,36h,48h,72h, and cells were counted to quantify the protein and the expression of the bFGF, the TGFβ1 was detected with western-blot. Results The expression of bFGF and TGFβ1 was positive in all groups,but the expression of growth factors was much more enhanced after stretching for 48h. Conclusion Mechanical stretch with 40% extend degree and one time but continues load can increase the expression of bFGF and TGFβ1.The peak time is 48h after stretching.
