摘要
目的 :建立一种机械作用细胞培养模型 ,将细胞培养在该模型上 ,验证模型的可行性。方法 :自行设计一种机械牵拉细胞培养模型 ,将 3~ 5代的皮肤成纤维细胞种植在弹性硅胶膜上 ,设计牵拉组与非牵拉组 (牵拉组采用 4 0 %的延长率 ) ,比较牵拉与非牵拉组 12h、 2 4h、 3 6h、 4 8h、 72h细胞数量 ,绘制生长曲线。结果 :细胞在牵拉模型上生长良好 ,4 0 %延长度的牵拉能刺激细胞增殖 ,生长曲线显示牵拉能明显刺激细胞增殖。结论 :细胞在牵拉模型上生长良好 ,牵拉能刺激细胞增殖 。
To establish a new mechanical strain unit for cell culture and verify its feasibility by using skin fibroblasts Methods:A mechanical strain unit was designed and manufactured in order to culture low pages skin fibroblasts on the elastic silicone membrances The experiment was carried out by using strained group and unstrained group The cell count was compared between two groups at the time points of 12h,24h,36h,48h,60h,72h,and the growth curves were drawn to see if there were any difference Results:The cultured skin fibroblasts of strained group grew better than the unstrained control And the cells were stimulated to proliferation when the elastic silicone membrane was strained to 40% stress rate Moreover the growth curve also shows the same effect Conclusion:The skin fibroblasts grow well in the mechanical strain unit And the mechanical stress can stimulate cells to proliferation The novel mechanical strain unit can be used to evaluate cell response sensitivity to mechanical stress in vitro
