摘要
利用BOX-PCR指纹图谱、结合16S rRNA全序列分析对分离自四川和重庆紫色土的38株硅酸盐细菌和2株参比菌株的遗传多样性和分类地位进行了研究.BOX-PCR指纹分析结果表明,尽管都是分离自紫色土的硅酸盐细菌,但是其遗传多样性明显,分别在54%相似性水平上分为4遗传群,约50%的供试菌株与胶胨样芽孢杆菌Bacillus mucilaginosusVKPM7519T聚在一群.对代表菌株K28和K38的16S rRNA全序列分析表明,K28与B.muculaginosus180D的16S rRNA全序列相似性为95.9%,而K38则与蜡质芽孢杆菌(Bacillus cereus)ATCC14579同源性高达99.9%,这是初次发现硅酸盐细菌在蜡质芽孢杆菌(B.cereus)中有分布.
Genetic diversity and phylogeny of 38 Silicate bacterial strains isolated from purple soils in Sichuan and Chongqing Provinces, together with two reference silicate bacteria strains, were analyzed by using BOX-PCR and 16S rRNA sequencing methods. The results of BOX-PCR showed that the genetic di- versity among silicate bacteria was existed, and all strains formed 4 genetic groups at the similarity of 54%, and there were more than 50% isolates from purple soil were close to B. mucilaginosus VKPM 7519T. Furthermore, 16S rRNA full sequence of K28 and K38 were performed, and the similarity of the 16S rRNA between Bacillus muculaginosus 180D and K28 was 95.9% ; however, 16S rRNA sequence ho- mology between B. cereus ATCC14579T and K38 was 99.9%, which was the first time to report that there was a silicate bacteria distributed in B. cereus.
