大花蕙兰外植体组培褐变的控制 被引量:11

摘要 对大花蕙兰离体繁殖中外植体的褐变进行了研究,结果表明:①假鳞茎切块和花茎切段褐变严重,选用茎尖褐变较轻。②在MS、改良MS和1/2MS三种培养基中,以无机盐浓度较低的1/2MS培养基、采用液体培养方式抑褐效果最好,但分化率差异不大,采用固体培养的材料,其分化率略高于液体培养。③在培养基中加入聚乙烯吡烙咯烷酮(PVP)、硫代硫酸钠、活性炭和维生素C,均能减轻褐变程度,其中以500mg/L PVP效果最好。④对褐变严重的外植体,进行接种前的抑褐剂预处理是有必要的,试验中以PVP(2g/L)和硫代硫酸钠(2g/L)预处理20min效果较好。 The browning of the explants in Cymbidium grandiflorium vitro propagation was studied, the results showed: (1)Browning of pseudobulb cuts and stem segments was serious while browning of shoot tip was lighter. (2)Among the three kinds of medium, such as MS, improved MS and 1/2 MS, inhibiting browning is best with 1/2 MS medium at low concentrations with inorganic salt and liquid cultivation, however, the difference of differentiation rate wasn't obvious, the differentiation rate by using solid training materials is slighdy higher than that by using liquid. (3)Browning could be reduced with the medium involved in polyvinylpyrrolidone (PVP),sodium thiosulfate, activated carbon and vitamin C, and the best effect was best with 500mg/L PVP. (4)It was necessary that the pretreatment of inhibiting browning to serious browning. The effect of pretreatment with PVP (2g/L) and sodium thiosulfate (2g/L) for 20min was best.
