摘要
目的探讨针刺对脑缺血再灌注损伤后星形胶质细胞增生的影响。方法建立大鼠大脑中动脉阻塞(MCAO)再灌注模型,随机分为对照组、模型组和针刺组,应用免疫组化和免疫印迹方法检测脑缺血再灌注后2d和7d损伤侧海马细胞周期素D1和胶质纤维酸性蛋白(GFAP)的表达。结果脑缺血再灌注后2d,损伤侧海马细胞周期素D1蛋白表达明显增强,与对照组比较差异有统计学意义(P〈0.01),脑缺血再灌注后7d,损伤侧海马GFAP的表达明显增强,与对照组比较差异有统计学意义(P〈0.01);给予针刺治疗后,细胞周期素D1蛋白和GFAP表达明显减弱,与模型组比较差异有统计学意义(P〈0.01)。结论针刺可抑制胶质细胞增生,从而有可能为神经再生和功能重建提供更有利的生存环境。其机制可能与其调控细胞周期有关。
Objective To investigate the effects of acupuncture on astrocyte proliferation in hippocampus of rats after cerebral isehemia-reperfusion. Methods Isehemia-reperfusion injury was induced by temporary middle cerebral artery occlusion (MCAO) in 36 Sprague-Dawley rats, who were divided randomly into a control group, a model group and an acupuncture group. The expression level of Cyclin D1 and glial fibrillary acidic protein (GFAP) was analyzed by immunoreactivity and Western blot. Results It was shown that the expression of Cyclin D1 and GFAP increased significantly (P 〈0. 01 ) at 2 and 7 days after cerebral ischemia-reperfusion, respectively. It was also shown that application of acupuncture could inhibit Cyclin D1 and GFAP (P 〈 0.01 ). Conclusion Acupuncture may inhibit glial proliferation by regulating cell cycle factor.
