摘要
目的:用五指山小型猪建立一种新的牙周感染合并动脉粥样硬化模型,探讨牙周炎与冠心病的相关性。方法:将4头小型猪随机分为三组:A单纯牙周感染模型组(n=1);B牙周感染合并动脉粥样硬化模型组(n=2),;C动脉粥样硬化模型组(n=1)。按各组实验设计相应处理后,喂养3~4个月,每20d检查牙周指标并测定体质量和血脂(TG、TC、HDL-C、LDL-C)水平。结果:牙周检查发现3个月后,A、B组牙周袋深度,龈沟出血指数较C组有明显增加;B、C组TC、HDL-C、LDL-C水平较A组明显升高,组织病理学检查发现主动脉形成早期动脉粥样硬化病变。结论:经过牙周检查、血脂检测和组织学检查和综合判定,初步成功建立了实验所需的动物复合模型,此模型可用于牙周炎与心血管疾病相关性的进一步研究。
AIM: To establish a new animal model of periodontal infection accompany with athen)sclerosis in minipigs. METHODS: Four 7 - month - old wuzhishan minipigs were divided into three groups ( A, B, C group). Group A: periodontal infection model was established by ligation,wounded,inoculated with P. gingivalis, which was fed with normal diet. Group B: atherosclerosis model was fed with high fat diet (HFD 15 g/Lcholesterol, 5 g/L cholate) ; Group C: periodontal infection and atherosclerosis model were got the same periodontal treatment as A, but fed with HFD. The total cholesterol (TC), triglyceride( TG), low density lipoprotein cholesterol (LDL- C), high density lipoprotein cholesterol( HDL- C)and periodontal indices were measured every twenty days. RESULTS: At the end of 3 months, the groups A and B showed deeper periodontal pockets and higher score of sulcus bleeding index (SBI) than C group. The serum lipoprotein levels of TC, LDL - C and HDI. - C were increased in groups B and C. The atherosclerotic pathological changes can be found in the aorta of groups B and C. CONCLUSION : After examination of periodontal status and serum lipoprotein level, the animal model of periodontal infection and atherosclerosis in minipigs was established successfully, which can be used in the study of the relationship between periodontitis and atherosclerosis.
