摘要
探索超声波辅助提取大豆卵磷脂的工艺条件,考察影响提取效果的主要因素,通过正交实验优化提取工艺。所得溶液用金属离子沉淀法进行纯化,用薄层层析法和紫外分光光度法对卵磷脂产品进行定性及定量分析。结果表明,最佳的工艺条件为乙醇浓度95%,萃取温度20℃、超声波萃取时间40 min、萃取次数为2次、固液比0.1 g/mL,金属离子沉淀剂加入量为15∶0(体积比)、pH值为9.0,在此条件下,产品的卵磷脂的含量可达75.3%,得率为27.63%。
The technics of ultrasound-associated extraction of phosphatidylcholine (PC) from soybean phospholipids were studied. The optimal conditions of the main factors were obtained by orthogonal method. At last, the solution was purified by precipitation with metal ion. The qualitative and quantitative analysis of different extracted PC fractions was carried out by TLC and ultraviolet spectrophotometry. Results presented demonstrate that the optimal conditions were concentration of ethanol 95%, extraction temperature 20 ℃, extraction time 40 min, 2 times and the ratio of liquid to material 1 g/mL. The amount of precipitator is 1:50 (v/ v) and pH9.0. Under this condition the PC purity is 75.3%, the yield is 27.63%.
