摘要
目的:利用正交试验研究三七总皂苷单体不同配伍对小鼠巨噬细胞源性泡沫细胞形成的影响,并评价最佳配伍的作用效果。方法:采用小鼠腹腔巨噬细胞与40mg.L-1氧化低密度脂蛋白孵育48h复制泡沫细胞模型,利用正交试验方法,以胆固醇酯(CE)含量为考察指标,对三七总皂苷中含量最高的3种单体成分的不同配伍影响泡沫细胞的形成进行观察。酶法测定细胞内总胆固醇(TC)、游离胆固醇(FC)、CE的含量及CE/TC的比值。选用最佳配伍组与三七总皂苷(80mg.L-1)的作用进行比较。结果:人参皂苷R1、Rg1、Rb1效应都显著,并且R1×Rb1和Rg1×Rb1的交互作用显著(P<0.01)。最佳单体配伍组合是R1∶Rg1∶Rb1=10-5∶10-5∶10-6mol.L-1。其作用效果与三七总皂苷(80mg.L-1)无显著差异。结论:该三七总皂苷单体配伍组合结果可证,可为临床提供理论依据。
OBJECTIVE: To observe the effects of different combinations of three monomers in saponins of Radix et Rhizorna Notoginseng on the formation of macrophage-derived foam cells in mice and evaluate the effect of the optimal combination of the monomers. METHODS: Macrophage cells from mice peritoneal were incubated with 40 mg·L^ -1 Ox-LDL for 48 h to establish foam cell model, and the effects of different combinations of the three monomers, which topping the list in terms of content in saponins of Radix et Rhizorna Notoginseng, on the formation of macrophage - derived foam cells in mice were observed with cholesterol ester content as index by orthogonal experiment. The cellular contents of total cholesterol (TC), free cholesterol (FC) and cholesterol ester (CE) and the ratio of CE/TC were detected by enzymic method. The effect of the best combination group of the monomers was evaluated and compared with that of saponins of Radix et Rhizorna Notoginseng(80 mg·L^-1) group. RESULTS: R1, Rg1 and Rb1 all showed remarkable effects on the content of CE in foam cell. The interaction of R1×Rb1 and that of Rg1× Rb1 were all remarkable (P〈 0.01). The best combination group was R1 : Rg1 : Rb1 = 10^-5 : 10 ^-5 : 10^ -6 mol · L^ -1. There was no significant difference between the best combination group of the monomers and saponins of Radix et Rhizorna Notoginseng(80 mg · L^-1) group in terms the effect on the content of CE in foam cell. CONCLUSION: The best combination of the monomers is confivmed,which provide theoretic basis for dinic.
