摘要
参照已发表的H5N1亚型禽流感的HA基因序列,设计了1对引物,对禽流感病毒新疆株A/Duck/XJ/3(H5N1)、A/Duck/XJ/4(H5N1)、A/CH/XJ/5(H5N1)、A/Goose/XJ/10(H5N1)和A/Duck/XJ/11(H5N1)进行RT—PCR扩增和克隆,获得了5个毒株的HA全长核苷酸序列,分别为1779bp、1774bp、1776bp、1758bp和1740bp序列分析表明,在HA切割位点处均有多个连续的碱性氨基酸(-R-E-R-R-R-K-K-R-)插入,具有高致病性禽流感病毒的特征,同源性分析表明,新疆分离株之间的核苷酸同源性在96.9%~98.8%;与来自青海湖斑头雁、广西天鹅、香港白鹭和台湾鸽等不同野禽毒株同源性为95.7%~100%;与来自香港、越南、浙江的人源禽流感毒株的同源性为95.2%~98.2%。
According to the published HA gene sequence of H5N1 subtype of avian influenza virus (AIV), a pair of primers was designed. The HA gene of A/Duck/XJ/3 (H5N1), A/Duck/XJ/4 (H5N1), A/ CH/XJ/5(H5N1), A/Goose/XJ/10(H5N1) and A/Duck/XJ/11(H5N1) were amplified by RT-PCR and cloned. The HA complete nucleotide sequences of each strain were 1 779 bp, 1 774 bp,1 776 bp,1 758 bp and 1 740 bp respectively. The sequences analysis indicated that there were serial basic amino acids (-R-E-R-R-R-K-K-R-) at the cleavage site of HA, which was consistent with the characteristics of highly patho- genic AIV. The sequences analysis showed that the nucleotide homology among Xinjiang AIV strains was 96. 9% ~ 98. 8%. Comparitive analysis of Xinjiang AIV strains with those from Qinghai, Guangxi, Hongkong and Tailand bird flu indicated that the nucleotide homology was 95.7%~100 %. The nucleotide homology of Xinjiang AIV strains in comparison with the human AIV isolates from Hongkong, Vietnam and Zhejiang was 95.2%~98.2%.
