摘要
用RT-PCR法,以1株H5N1亚型禽流感病毒新疆分离株A/Duck/XJ/4为模板,扩增了NP全基因cD-NA,克隆至T载体,并进行序列测定。序列分析结果表明:扩增的NP基因全长1518 bp,最大的开放阅读框在18~1514 bp,编码499个氨基酸。将A/Duck/XJ/4毒株NP基因序列与15株A型流感病毒NP基因序列进行比较,各毒株间NP基因核苷酸序列同源性81.4%~99.0%,编码的氨基酸同源性在92.8%~99.6%。
NP gene of A/Duck/XJ/4 strain of N5H1 avian influenza virus was used as template. The cDNA of NP gene was amplified by RT-PCR method and was cloned into T vector and then was sequenced. The cloned 1 518 bp fragment covered open reading frame of NP gene and encoded 499 amino acids. The results of sequence analysis indicated that the nucleotide sequence of NP gene of A/Duck/XJ/4 strain shared 81.4%--99.0% homology with other fifteen isolates and encoded amino acid sequence shared 92.8%--99.6% homology with other fifteen isolates.
