Expression of Livin and Caspase-3 proteins in human non-small cell lung carcinoma tissue

摘要 目的:检测人非小细胞肺癌(NSCLC)组织中Livin和Caspase-3蛋白的表达情况,并探讨2者在NSCLC发生、发展中的作用。方法:采用免疫组织化学法分别检测70例NSCLC组织(男42例,女28例;鳞状细胞癌50例(高分化12例,中分化18例,低分化20例),腺癌20例(高分化6例,中分化5例,低分化9例);有淋巴结转移46例,无淋巴结转移24例)、40例癌旁组织、30例正常肺组织中Livin和Caspase-3蛋白的表达水平,并分析2者与肺癌的组织类型、分化程度、淋巴结转移的关系,以及2种蛋白表达的相关性。结果:NSCLC组织中Livin蛋白的阳性表达率(85.7%)高于癌旁组织的45.0%和正常肺组织的0%(P均<0.05);Livin蛋白的阳性表达率与NSCLC的分化程度、组织学类型无关(P均>0.05),与淋巴结转移情况有关(P<0.05)。NSCLC组织中Caspase-3蛋白的阳性表达率40.0%低于癌旁组织的72.5%和正常肺组织的83.3%(P均<0.05);Caspase-3蛋白的阳性表达率与NSCLC组织的分化程度、淋巴结转移情况有关(P均<0.05),与组织学类型无关(P>0.05)。NSCLC组织中Livin与Caspase-3蛋白的表达呈负相关(r=-0.299,P<0.05)。结论:Livin蛋白可能通过抑制Caspase-3蛋白的活性抑制细胞凋亡和促进细胞增殖,加速细胞的恶性转化和NSCLC的发生。 Aim : To detect the expression of Livin and Caspase-3 proteins in non-small cell lung carcinoma (NSCLC) tissues and their relationship with the clinical characteristic. Methods:The expression of Livin and Caspase-3 proteins were detected by immunohistochemistry method in 70 cases of NSCLC (42 cases of male, 28 cases of female; 50 cases of squamous cancer( 12 cases of well-differentiated, 18 cases of moderate-differentiated,20 cases of poorly-differentiated), 20 cases of adenocarcinoma(6 cases of well-differentiated,5 cases of moderate-differentiated, 9 cases of poorly-differentiated) ; 46 cases with lymph node metastasis,24 cases without lymph node metastasis) and 40 pericarcinomatous tissues, 30 normal lung tissues. Expression of these proteins were analyzed for correlation with histological typing, degree of differentiation and lymph node metastasis. Results: The positive expression rate of Livin protein in NSCLC (85.7%) was higher than those in periearcinomatous lung tissue (45.0%) and normal lung tissue (0%) (P 〈 0.05 ). The positive expression rate of Livin protein in NSCLC tissue was related with lymph node metastasis ( P 〈 0.05 ) , except that differentiation degree and histological, type (P 〉 0.05 ). The positive expression rate of Caspase-3 protein in NSCLC (40.0%) was lower than those in periearcinomatous lung tissue(72.5% ) and normal lung tissue (83.3%) (P 〈 0.05 ). The positive expression rate of Caspase-3 protein in NSCLC tissue was related with differentiation degree and lymph node metastasis ( P 〈 0.05 ) , except that histological type (P 〉 0.05 ). The expression of Livin protein was negatively related to that of Caspase-3 protein in NSCLC tissues (r = - 0. 299,P 〈 0.05 ). Conclusion : The Livin protein may promote cell transformation and carcinogenesis of NSCLC through inhibiting Caspase-3 activity and apoptosis and increasing cell proliferation.
