摘要
目的探讨罗格列酮(RGZ)对裸小鼠移植瘤过氧化物酶体增殖物激素受体-γ(PPAR-γ)表达的影响。方法采用不同浓度和剂量的RGZ对荷瘤裸小鼠进行灌胃干预3周后,RT-PCR实时荧光定量检测PPAR-γ的△Ct值和表达拷贝数,测量各组瘤体体积(TV)和重量(TW),计算相对肿瘤体积(RTV)和相对肿瘤增值率(T/C%)。结果胆管癌移植瘤随RGZ剂量增高PPAR-γ的表达升高,而相对肿瘤增值率T/C%减少;高剂量组与对照组比较差异显著(P<0.001)。结论在裸小鼠移植瘤内RGZ能呈剂量依赖性地上调PPAR-γ表达。
Objective To study the effect of RGZ on expression of PPAR-γ in transplantation tumor of Cholangiocarcinoma. Methods To interfere the nude mice bearing cancer of Cholangiocarcinoma with RGZ in different concentrations and dosages, animals were sacrificed 3 week after intragastric administration. To measure tumor mass and weights, to evaluate tumor size, tumor increasing ratio T/C% and to detect the expression of PPAR-γ by RTPCR by Real Time Fluorescent Quantitation, statistic analysis as well. Results Following the dose of RGZ raising the expression of PPAR-γ while the tumor size, weight, the relative tumor growth ratio T/C% decrease in the transplantation tumor of Cholangiocarcinoma. Statistic treatments showed no significance between the control and low dose groups ( P 〉 0. 05 ) ; but as comparing with high dose group there is obvious significance (P 〈 0. 001 ). Conclusion In vivo, RGZ shows obvious effect of up-regulation PPAR-γ expression in transplantation tumor of Cholangiocarcinoma.
