摘要
目的建立HPLC梯度洗脱法测定天山花楸叶与果中绿原酸含量的方法。方法采用ZORBAS ODS C18(4.6mm×15cm,5μm)色谱柱,甲醇-0.02%磷酸溶液为流动相(梯度洗脱),流速为1.0ml·min^-1,检测波长为326nm。结果绿原酸线性范围为3.952~19.76μg/ml,叶中绿原酸含量1.982%(RSD=0.58%,n=3),平均回收率为99.42%;果中绿原酸含量2.259%(RSD=1.2%,n=3),平均回收率为99.32。结论该方法经济、简便、快速、准确、可靠,可用于该药材的质量控制。
Objective To establish methods for determination of chlorogenic acid in Sorbus tianschanica. Methods The HPLC system consisted of the column of ODS ZORBAS ODS (4.6 mm × 15 cm,5 μm) column and MeOH -0.02% H3 PO3 as the mobile phase. The flow rate was 1.0 ml · min^-1. The detection wavelength was 326 nm. Results The method was linear within the range of 3. 952 - 19.76 μg/ml. Chlorogenie acid in the leaf of S. tianschanica was 1. 982% (RSD% = 0.58, n = 3 ) and the average recovery was 99.42%. Chlorogenie acid in the fruit of S. tianschanica was 2. 259% ( RSD% = 1.2, n = 3 ) and the average recovery was 99.32%. Conclusion The method is simple, sensitive, quick and highly reproductive and it is suitable for analysis of chlorogenie acid in S. tianschanica.
