摘要
目的:明确双黄连粉针剂高剂量给药时有效成分绿原酸在大鼠体内的动态变化规律及其在唾液中的分布情况。方法:大鼠740 mg.kg-1静脉注射双黄连粉针后,反相高效液相色谱法测定绿原酸在血液、唾液中的浓度,采用Multi97进行统计处理,确定药动学参数。结果:绿原酸在大鼠体内符合二室模型分布,其主要药动学参数如下:t1/2=0.63 h,AUC=62.40μg.h-1.mL-1,Vc=3.58 L,CL=4.03 L.h-1。绿原酸的唾液浓度范围为:(0.42~9.88)μg.mL-1。结论:双黄连粉针高剂量给药时绿原酸呈非线性药动学消除过程,在唾液中的分布浓度较低。
