替米沙坦对高糖高脂喂养大鼠一氧化氮和一氧化氮合酶的影响 被引量:2

摘要 目的观察替米沙坦对高糖、高脂喂养大鼠血清一氧化氮(NO)和内皮型一氧化氮合酶(eNOS)的影响。方法30只SD大鼠随机分为空白对照组、模型组和用药组,模型组和用药组给予高糖、高脂饲料喂养,用药组同时给予替米沙坦6mg·kg-1·d-1灌胃10周。实验结束时测定NO、eNOS含量,取大鼠主动脉弓电镜观察。结果模型组与空白对照组比较,NO明显降低(30.50±5.40和39.34±7.34,P<0.01),eNOS明显降低(35.07±6.59和47.07±6.56,P<0.01),电镜下可观察到内膜破损、内皮细胞脱落;用药组与模型组比较,NO升高(37.42±5.75和30.50±5.40,P<0.05),eNOS明显升高(44.81±7.00和35.07±6.59,P<0.01),电镜下可观察到内膜完整接近空白对照组。结论替米沙坦可使高糖、高脂喂养大鼠NO和eNOS含量升高。 Objective Observe the effect of telmisartan on NO and eNOS of the rats fed a high-sucrose and high-fat diet. Methods Thirty SD rats were randomly divided into control group, model group and drug group, rats of the model group and drug group were fed a high-sucrose and high-fat diet, rats of the drug group were treated with the intragastric infusin of telmisartan 6 mg·kg^-1·d^-1 for ten weeks. At the end of the trial, sampiing blood and measuring the concentrations of nitric oxide (NO) and endothelial nitric oxide synthase(eNOS), aortic arch was examined through transmission electron microscopy. Results Compared with the control group, NO was significantly decreased (30.50±5.40 vs 39.34±7.34,P〈0,01 ), eNOS was significantly decreased (35.07±6.59 vs 47.07±6.56,P〈0.01 ), endomembrane was not continuous and an endothelial cell was falling off in the model group; compared with the model group, NO was increased (37.42±5.75 vs 30.50±5.40,P〈0.05), eNOS was significantly increased (44.81±7.00 vs 35.07±6.59,P〈0.01),endomembrane was continuous in the drug group. Conclusion Telmisartan can increase NO and eNOS of the rats fed a high-sucrose and high-fat diet.
