摘要 目的通过检测2型糖尿病大鼠尿液代谢谱的变化,探讨代谢组学在糖尿病研究中的应用。方法 SD大鼠高糖高脂饲料喂养6周后,腹腔注射链脲菌素(Streptozotocin,STZ)37 mg/kg建立2型糖尿病模型,动态检测空腹血糖(FBG)变化,检测甘油三酯(TG)、总胆固醇(TC)、游离脂肪酸(FFA)及胰岛素(INS)水平。分别于造模前、造模后1、2、4、6、8周收集大鼠尿液,采用氯甲酸乙酯(ECF)衍生和气相色谱质谱联用(GC/MS)的方法检测大鼠尿液代谢谱的变化。结果造模后大鼠FBG持续升高,FFA、TG、TC明显升高,血清INS含量明显降低;造模后,模型组大鼠代谢谱与对照组完全区分,并随时间变化逐渐偏离;比较两组差异,从差异变量中鉴定出10个生物学标记物。结论高糖高脂喂养联合小剂量STZ造模2型糖尿病大鼠尿液代谢组学发生明显变化,尿液代谢组学在一定程度上反映2型糖尿病大鼠的病理变化。 Objective To investigate the urinary metabonomic variation in diabetic rats in order to explore the ap-plication of metabonomics in research of diabetes.Methods Male Sprague-Dawley rats were fed with high-fat and high-su-crose diet for 6 weeks and then injected with vehicle or streptozotocin(STZ) in a dose of 37 mg /kg.Fasting blood glucose(FBG) was monitored weekly after STZ injection.Urines of normal rats and model rats were collected at 1,2,4,6,8 weeks after STZ injection.Serum insulin(INS),triglyceride(TG),total cholesterol(TC) and free fatty acids(FFA) were measured at 8 weeks after STZ injection and the rats were sacrificed.The metabolic profiles were analyzed using ethyl chloroformate(ECF) derivatization and gas chromatography-mass spectrometry(GC /MS).Results Compared with the control rats,the FBG,TG,FFA were significantly higher while the INS was reduced in the model rats.The metabolism profiles of the model rats were significantly different from that of the control rats,and changed with time.Comparative study was applied and ten biological markers were different between the normal and model rats.Conclusions Significant chan-ges in urine metabonomics occurre in type 2 diabetic rats induced by high-fat and high-sucrose diet and low dose of STZ,and it may reflect to a certain extent the pathological changes of type 2 diabetic rats.
