实验性阻塞型睡眠呼吸暂停综合征小型猪的血浆同型半胱氨酸水平 被引量:4

摘要 目的利用动物模型观察实验型阻塞型睡眠呼吸暂停综合征小型猪的血浆同型半胱氨酸(Hcy)水平,以探讨睡眠呼吸暂停致动脉粥样硬化的可能机制。方法中国小型猪16头,分为阻塞型睡眠呼吸暂停组(OSA组)和对照组,每组8头。OSA组用凝胶注射法制作动物模型。分别于制作模型前及12周后取实验动物静脉血检测血浆Hcy、叶酸及维生素B12水平。结果(1)造模后,OSA组Hcy水平明显升高,前后分别为(8.94±2.43)和(12.25±1.44)μmol/L,两者相比差异有统计学意义(P≤0.05),对照组Hcy水平无明显变化。造模后OSA组与对照组Hcy值相比差异有统计学意义(P<0.05)。(2)两组动物叶酸及维生素B12水平实验后与实验前相比无明显变化。结论实验性OSA使机体血浆中Hcy水平异常升高,促进机体动脉粥样硬化的发生和发展。 Objective To explore the possible mechanism of sleep apnea in causing atherosclerosis through observing plasma homocysteine levels in the animal models with experimental obstructive sleep apnea syndrome. Methods Sixteen Chinese miniature pigs were divided into obstructive sleep apnea (OSA) group and control group, with 8 pigs in each group. Animal models were made by gel injection method in OSA group. Before injection and 12 weeks after injection, venous blood was obtained from experimental animals to measure homocysteine (Hcy), folic acid(Fol) and vitamin Blz (VBIz). Results (1) After gel injection, the level of Hcy was obviously elevated in OSA group, increasing from(8.94±2.43)μmol/L to( 12.25 ± 1.44)μmol/L. Significant difference existed in the Hcy level between before and after gel injection (P〈0.05). No significant change in the level of Hcy was found in control group. There was significant difference in the Hcy level between OSA and control groups (P〈0.05). (2) In both groups, there were no significant changes in the levels of Fol and VB12 before and after gel injection. Conclusion Experimental obstructive sleep apnea can increase the level of plasma homocysteine, which results in endothelial injury and dysfunction, and promotes the initiation and development of atherosclerosis.
