摘要
目的观察肿瘤坏死因子在大鼠脑出血后的变化及其与脑水肿的关系。方法采用随机方法将40只Sprague-Dawley实验大鼠分为对照组、脑出血模型组,每组又按照不同的时相点6、24、72、168h分为4个亚组,每个亚组5只。采用自体血脑内注入法制作大鼠脑出血模型,对照组只进针不注血。干、湿重法检测脑含水量;酶联免疫吸附法(ELISA)测定血浆肿瘤坏死因子-α(tumour necrosis factor-alpha,TNF-α)的浓度。结果脑出血模型组大鼠各个时相点脑含水量、血浆TNF-α的浓度均显著高于对照组(P<0.01)。结论脑出血后大鼠血浆肿瘤坏死因子-α浓度明显增高。脑出血后脑水肿的产生可能与肿瘤坏死因子的大量释放有关。
Objective To observe the change of turnout necrosis factor-alpha(TNF-α) and its relations with brain edema in rats following intracerebral hemorrhage( ICH ). Methods Fouty Sprague-Dawley rats were divided into 2 groups at random: control group and ICH model group, There were 20 rats in each group. ICH model was induced by infusion of autologous whole blood into the caudate-putamen. Rats in control group were treated same as above, but no blood injected. Brain water content was measured by way of dry-wet weight method; TNF-α was measured by ELISA. Results Compared with control group, brain water content and TNF-α in plasma were obviously higher in ICH model group (P〈0.01). Conclusion TNF-α content in plasma greatly increases after ICH. The development of brain edema are probably connected with TNF-α's releasing after intracerebral hemorrhage in rats.
