摘要
目的观察甘草酸二胺对大鼠脑出血(Intracerebra lhemorrhage,ICH)后脑水肿的疗效。方法参照Rosenberg等报道的方法制备大鼠脑出血模型,术后24h对大鼠进行头颅磁共振(Magnetic resonance,MR)扫描,判断脑出血模型制作是否成功,将模型制作成功大鼠随机分为脑出血模型组、甘草酸二胺治疗组,另设假手术组,所有大鼠第5天MR扫描后处死,采用干湿重法测定大鼠脑组织含水量,用ELISA法测定脑组织中肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、白介素-6(IL-6)的表达水平。结果脑出血模型组大鼠脑含水量、脑组织中TNF-α、IL-1β、IL-6水平明显高于假手术组,甘草酸二胺治疗组脑含水量、TNF-α、IL-1β、IL-6较模型组明显下降(P<0.01)。磁共振扫描显示模型组血肿血肿周围水肿范围明显增大,甘草酸二胺治疗组血肿周围未见明显水肿。结论甘草酸二胺能减轻大鼠脑出血后脑水肿,其机制可能与减轻炎症反应有关。
Objective To observe the efficacy of diammonium glycyrrhizinate t in the treatment of cerebral edema after intracerebral hemorrhage(ICH) in rats.Methods 24 hours after the ICH model established,MR scanning was performed.The successful models were randomly divided into 2 groups:ICH model group and treated group,and sham-operated group was built up.5 days after operation,MR scanning was performed firstly,and then the rats were sacrificed to measure the brain water content with wet-dry weight method.The contents of TNF-α,IL-1β,IL-6 in the brain tissue were detected with ELISA method.Results The contents of TNF-α,IL-1β,IL-6 and the water content were reduced in the treated group as compared to ICH group.Perihematoma edema was not found in treated group,but the scope of edema around the hematoma in ICH model group increased.Conclusion Diammonium glycyrrhizinate could reduce cerebral edema after ICH in rats,whose mechanisms may be related to resultant alleviation of cerebral inflammatory.
