摘要
目的探讨胰腺癌中组织因子途径抑制物-2(TFPI-2)的表达及作用。方法采用免疫组织化学方法检测41例胰腺癌组织中TFPI-2,Ki-67和VEGF的表达及CD34单克隆抗体标记的微血管密度(MVD)。结果Ⅰ,Ⅱ期胰腺癌TFPI-2阳性表达率(17/26)高于Ⅲ,Ⅳ期(2/15),高分化癌(12/19)高于中、低分化(7/22)(均P<0.05);Ⅰ,Ⅱ期癌细胞增殖指数(PI)(24.45±6.19)低于Ⅲ,Ⅳ期癌(30.84±6.70),高分化癌(24.42±6.29)低于中、低分化癌(28.84±7.12),(均为P<0.05);Ⅰ,Ⅱ期癌VEGF的阳性表达率(16/26)低于Ⅲ,Ⅳ期癌(14/15),高分化癌(11/19)低于中、低分化癌(19/22)(均P<0.05);Ⅰ,Ⅱ期癌MVD值(26.92±1.06)低于Ⅲ,Ⅳ期癌(28.47±2.23),高分化癌(19.00±2.58)低于中、低分化癌(16.68±1.79)(均P<0.05)。胰腺癌组织中TFPI-2的表达与Ki-67和VEGF的表达呈负相关(r=-0.62,P<0.05;r=-0.54,P<0.05),MVD与TFPI-2表达亦有相关。结论TFPI-2表达能够抑制胰腺癌细胞增殖,并通过下调VEGF的表达抑制肿瘤新生血管的形成,这可能是其抑制胰腺癌的生长、转移的重要机制。
Objective To explore the expression and action of tissue factor pathway inhibitor 2 ( TFPI-2 ) in pancreatic carcinoma ( PC ). Methods Tissue specimens from 41 pancreatic cancers were used to detect the expression of TFPI-2, Ki-67, VEGF and MVD tagged with CD34 by immunohistochemistry. Results The positive expression of TFPI-2 in stage Ⅰ and ⅡPC (17/26) was higher than that in stage Ⅲand Ⅳ(2/15) PC. The positive expression of TFPI-2 in well-differentiated PC ( 12/19 ) was also higher than that in moderateand poorly-differentiated PC (7/22) ( P 〈 0.05 ) . The cell proliferation index in stage Ⅰ, Ⅱ(24.45 ±6. 19) and well-differentiated PC (24.42 ±6. 29) was lower than that in stage III , IV (30.84±6. 70) and moderate-, and poorly-differentiated PC (28. 84 ±7. 12) (P 〈 0. 05). The positive expression of VEGF in stage Ⅰand ⅡPC ( 16/26 ) was higher than that in stage Ⅲand Ⅳ(14/15). The positive expression of VEGF in well-differentiated PC (11/19) was also higher than that in moderate- and poorly-differentiated PC (19/22) (P 〈 0.05) . The value of MVD in stage Ⅰ, Ⅱ(26.92 ±1. 06 ) and well-differentiated PC ( 19. 00 ±2. 58 ) was lower than that in stage of Ⅲ, Ⅳ(28.47±2.23) and moderate- and poorly-differentiated PC (16. 68 ±1. 79), (P 〈 0. 05 ). The expression of TFPI-2, Ki-67 and VEGF was negatively correlated. The MVD was related to the expression of TFPI-2. Conclusions TFPI-2 can inhibit cell proliferation of PC, and by reducing the expression of VEGF can inhibit the angiogenesis of new vessels in PC, which may play an important part in inhibitory effect on invasion and metastasis of PC.
