摘要
通过HPLC确定灌胃芦荟药物在家兔血清中达到峰值时相,确定最佳采血时间,制备芦荟药物血清,将血清加入糖基化终产物(AGEs)作用过的肾系膜细胞(RGMC)中,孵育24 h。之后,检测细胞内谷胱甘肽(GSH)和氨基葡萄糖苷酶(NAG)含量,超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、山梨醇脱氢酶(SDH)和醛糖还原酶(AR)的活性,研究芦荟药物血清对RGMC的保护效果,初步探讨芦荟防治糖尿病的机理。结果表明,芦荟药物在家兔灌胃后2 h的血清中血液达到相对峰值;AGEs作用过的细胞经芦荟药物血清处理后,其GSH含量、SOD活性均较未加芦荟处理的A组有所升高,且差异极显著(P<0.01),其SDH和AR的活性也较未经芦荟处理的A组极显著下降(P<0.01)。表明芦荟药物血清对AGEs损伤后的RGMC细胞有一定的保护修复作用,芦荟保护RGMC细胞的机制可能是通过改善细胞的氧化应激状态,同时降低多元醇通路的2种关键酶的活性而起作用的。
To investigate the protective effect of aloe medicated serum on RGMC and to probe into the mechanism of the curing effect of aloe on diabetes, the peak time of the aloe content in rabbit serum was determined by HPLC. Serum was obtained from the blood after aloe garage,then medicated serum was added into the cells treated by AGEs. After 24 h incubation,indexes such as GSH ,NAG content, SOD, CAT, SDH and AR activities in cells were measured. The peak time of aloe content in rabbit serum was 2 hours after gavage;GSH content and SOD activity were markedly higher (P(0.01)in the medicated serum treated AGEs group than that in untreated AGEs group,while AR and SDH activity decreased in the medicated serum treated with AGEs group. It can be concluded that Aloe medicated serum exerted protective effect on ceils treated with AGEs,and it can be hypothesized that therapeutical effect of aloe on diabetes and its related complications was probably due to alleviation of the oxidative stress and decrease in the activities of two key enzymes in the polyol pathway in ceils.
