摘要
目的检测组织蛋白酶B(cathepsin B,CB)与基质金属蛋白酶9(matrix metalloproteinase-9,MMP-9)在人类上皮性卵巢肿瘤中的表达,探讨两者与卵巢癌细胞的分化增殖、侵袭转移的关系。方法采用链霉素亲和生物素-过氧化物酶法(SP)进行免疫组织化学染色,用CB抗体、MMP-9抗体标记上皮性卵巢肿瘤组织标本,综合染色强度和阳性细胞占总体细胞数的百分比进行染色判定,结合有关病理特征进行分析。结果经统计学分析,CB、MMP-9在上皮性卵巢癌中的表达与年龄、病理类型无相关性(P>0.05),而与卵巢癌的临床分期、有无转移及腹水呈正相关(P<0.05);CB与卵巢癌的组织学分级呈正相关(P<0.05);上皮性卵巢癌中CB和MMP-9表达呈正相关(r=0.652,P<0.05)。结论CB、MMP-9与卵巢癌的恶性生物学行为密切相关,两者在其侵袭转移过程中发挥了重要的协同作用。
Objective To study the expression of CB and MMP-9 in ovarian tumor with the histodifferentiation and invasion and metastasis. Methods The expressions of CB and MMP-9 were detected by S-P immunohistochemistry method in histological section of ovarian tumor, and the relationships between the results and the clinical pathological parameters of the patients were assessed. Results The expression of CB and MMP-9 had marked correlation (P〈0.05) with clinical stage, metastasis and ascites of the ovarian cancer. Moreover, the expression of CB had marked correlation (P〈0.05) with pathologic grade. The positive correlation was observed between the expression of CB and MMP-9 (r=0.652, P〈0.05). Conclusion The expression of CB and MMP-9 in ovarian cancer cells is closely related to the tumor malignancy biological behaviour of ovarian cancer. They play an important synergistic action in the invasion and metastasis of ovarian cancer.
