摘要
目的观察白藜芦醇(resveratrol,Res)在体外对胃癌细胞系BGC823增殖及其基质金属蛋白酶-9(matrix met-alloproteinase-9,MMP-9)表达的影响,初步探讨Res抑制胃癌细胞BGC823侵袭转移的机制。方法应用不同浓度(10、50、100μmol/L)的Res分别作用于人胃癌细胞系BGC823细胞24、48及72 h,采用四甲基偶氮唑盐微量酶反应比色法(methyl thiazolyl tetrazolium,MTT)观察Res对BGC823细胞增殖的影响,应用反转录聚合酶链式反应(reverse transcription polymerase chain reaction,RT-PCR)检测MMP-9 mRNA的表达,应用蛋白印迹分析(Westernblot)法检测MMP-9蛋白表达情况。结果 Res对胃癌BGC823细胞的增殖有明显的抑制作用,且呈剂量和时效关系。与对照组相比,Res作用BGC823细胞后,MMP-9 mRNA及蛋白表达水平均明显下降(P<0.05)。结论 Res对胃癌细胞的生长具有抑制作用,且能下调其MMP-9的表达,可能具有抑制胃癌细胞侵袭转移的作用。
Objective To observe the effects of resveratrol on proliferation of human gastric carcinoma cell line BGC823 cells and expression of matrix metalloproteinase-9(MMP-9) in vitro so as to explore the mechanism of resveratrol's inhibiting the metastasis of BGC823 cells.Methods BGC823 cells were treated with different concentrations of resveratrol(10,50 and 100 μmol/L)for 24,48 and 72 h,respectively.The effects of resveratrol on proliferation of BGC823 cells were assessed with methyl thiazolyl tetrazolium.The expression of MMP-9 mRNA was determined by reverse transcription polymerase chain reaction while MMP-9 protein was identified by Western blot analysis.Results Resveratrol could inhibit the proliferation of BGC823 cells in dose-and time-dependent manners.Moreover,compared with those in control group,both MMP-9 mRNA and protein expressions were markedly reduced after resveratrol treatment(P〈0.05).Conclusion Resveratrol can inhibit the proliferation of BGC823 cells and down-regulate MMP-9 expression.Therefore,resveratrol may suppress the invasion and metastasis of gastric carcinoma.
