The polyhedrin gene (Ph) of Helicoverpa (Heliothis) armigera,a singlenucleocapsid nuclear polyhedrosis virus (HaSNPV) Hubei isolate located within a 4.2kb SalⅠ fragment,was cloned using BmNPV Ph gene as α- 32 P-labeled probe and Southern hybridization method.1701 nucleotides (nt) of the genome containing the Ph was sequenced,and an open reading frame (ORF) of 738nt encoded a protein of 246 amino acids with a MW of 29kD was predicted.With canonical ATAAG promoter motif,a conserved 14-nt sequence (Rohrmann box) AAAATAAGTATTTT associated with transcriptional start sites in other polyhedrin genes was identified at -48 to -52 nt upstream from the start codon.A putative polyadenylation signal AATAAA,was found at 258 nt downstream of the polypeptide translational stop codon.Comparison of the nucleotide sequence and deduced amino acid of HaSNPV Ph with other NPV polyhedrins and granulosis virus (GV) granulins showed a high degree of homology with 20 other NPVs,particularly closely related to that of HzSNPV (97%),and most distantly related to that of Neodiprion sertifer SNPV(44.5%).
