摘要
采用稀盐法对马尾松毛虫蛹中的蛋白质进行了提取研究。结果表明,盐浓度、料液比、提取pH、提取温度、提取时间和沉淀pH对蛹蛋白质的提取率有显著影响(P<0.05),而沉淀温度和沉淀时间对提取率无显著影响。实验确定的最佳条件为盐浓度1.5%、料液比为1∶15(w/v)、提取时间为120min、提取温度为40℃、提取液pH11、沉淀pH为4.5、沉淀温度为25℃、沉淀时间为30min。在此条件下,所得蛋白质产品的粗蛋白含量为91.3%,总氮提取率为80.21%。首次提出了从害虫松毛虫蛹中提取蛋白质的可行技术。
The protein extraction from pupae of DendroUmus punctatus walker with salt method was investigated. The results showed that salt concentration, relative amount of materials and extraction solvent, extraction pH, extraction temperature, extraction time and precipitation pH showed significant effects on the extraction rate of the protein(P〈0.05), while precipitation temperature and precipitation time had no significant effects on it. The optimal condition was as follows. Salt concentration was 1.5%, relative amount of materials and extraction solvent was 1:15(w/v), extraction pH was 11.0, extraction time was 120min, extraction temperature was 40℃, precipitation pH was 4.5, precipitation temperature was 25℃, precipitation time was 30min. The protein content of the product obtained at the optimal condition was 91.3%, and the extraction rate was 80.21%. This work firstly reported a feasible technology for protein extraction from pupae of Dendrolimus punctatus walker with salt method.
