Effect of estrogen on the expression of pigment epithelium derived factor in cultured retinal Müller cells under the anoxic condition

摘要 目的探讨雌激素对缺氧视网膜Müller细胞色素上皮衍生因子(PEDF)表达的影响。方法采用逆转录聚合酶链反应(RT-PCR)和Western blotting印迹分析方法分别测定不同浓度(10^-7、10^-6、10^-5mmol/L)雌二醇(E2)作用于缺氧Müller细胞后,细胞内PEDFmRNA及蛋白表达水平。结果缺氧24h后PEDFmRNA及蛋白表达明显降低,10^-5、10^-6mmol/L E2作用于Müller细胞后可明显缓解由于缺氧引起的细胞内PEDFmRNA及蛋白表达的降低,并与E2的浓度有关。结论雌激素可以调控PEDF的表达,其可能在雌激素对视网膜新生血管形成的调控中起重要作用。 Objective To observe the effect of estrogen on the expression of pigment epithelium derived factor (PEDF) in cultured retinal Müller cells under the anoxic condition. Methods After the anoxic retinal Müller cells were treated with estrogen (E2) with the concentration of 10^-5, 10^-6, and 10^-7 mmool/L, the level of expression of PEDF mRNA and the protein was detected by reverse transcriptionpolymerase chain reaction and Western blotting analysis. Results The expression of PEDF mRNA and protein decreased 24 hours after anoxia. E2 with the concentration of 10^-5 and 10^-6 mmol/L inhibited the decrease of expression of PEDF mRNA and protein induced by anoxia, which related to the concentration of E2. Conclusion strogen can regulate the expression of PEDF, which may play an important role in the regulation of retinal neovascularization.
