废用性骨质疏松动物承重骨的骨改建活动和力学性能变化的研究 被引量:10

摘要 目的:探讨构建废用性骨质疏松动物模型的方法,及废用性骨质疏松发展过程中不同时间内骨的结构、矿物质含量以及受其影响的骨的生物力学性能的变化情况。方法:选用雄性日本大耳白兔55只,随机石膏固定50只兔的右后肢作为实验侧,左侧后肢作为自身对照侧,另5只作为空白对照组饲养3个月。固定后2、4、6、8、12周各处死10只兔。取双侧胫腓骨行力学试验,测定钙元素的含量。取双侧跖骨头,进行HE染色和Masson染色,组织学观察。结果:对于最大负荷、弹性模量和骨钙含量等指标,实验侧胫腓骨在不同固定时间的差别,实验侧和对照侧间的差别均有统计学意义(P<0.05);对照侧和空白对照组间,对照侧间的差别无统计学意义(P>0.05)。实验侧骨皮质变薄,孔隙增多,骨小梁稀疏、纤细并有多处断裂,小梁间吻合减少,骺板下区空旷,髓腔扩大,破骨细胞数增多。Masson染色见随着废用时间的延长,新生胶原的量也在减少。结论:石膏固定实验动物肢体,限制其负重,是有效的构建废用性骨质疏松动物模型的方法。在废用性骨质疏松形成过程中,骨钙含量、骨的整体生物力学性能持续下降,但速度趋缓,以适应所处的应力环境和功能状态。 Objective: To study the changes of the bone structure, bone mineral content and bone biomechanical properties in the development of the disused osteoporosis. Methods: 55 male Japanese rabbits were used in our research. 50 rabbits were selected as trial group and the other 5 as control group fed 3 months. Their right hind limbs were immobilized using cast as test sides, the left ones as control sides corresponded. 2,4,6,8,12 weeks after fixation, 10 rabbits were sacrificed. The tibiofibula were tested using the mechanical test machine and then the content of calcium was measured. The metatarsus were decalcified and then stained with HE and Masson stain. Results: The differences of maximum load(ML), Yong' s modulus(YM) and content of calcium (Ca) of the tibiofibula in test side during different stages were significant statically (P〈0.05). The differences of ML, YM and Ca of the tibiofibula between test side and control side were significant statically (P〈0.05). The differences of ML(P〉0.05), YM (P〉0.05) and Ca (P〉0.05) of the tibiofibula between control side and blank control group were not significant statically. The wall of cortical bone became narrower and the trabucular bone became thinner. Conclusion: The content of calcium and whole mechanical properties of the bone decrease constantly during the development of the disused osteoporosis, but the trend of decline becomes slowly. Our results illuminate that the metastasis of bone structure is adapting to the stress environment during the immobilization. That newly formed collagen kept decreasing maybe correspond to the decreasing of bone remodeling.
