载药磷酸钙骨水泥促进骨质疏松大鼠骨折愈合的研究 被引量:1

摘要 目的 研究磷酸钙骨水泥(CalciumPhosphateCement,CPC)与重组人骨形态发生蛋白- 2 (RecombinantHumanBoneMorphogeneticProtein -2 ,rhBMP -2 )复合材料对骨质疏松大鼠骨折愈合的影响。方法 取4月龄雌性大鼠6 0只,双侧卵巢切除后形成骨质疏松动物模型。然后制作右侧股骨远端松质骨骨折,将上述动物随机分成3组,每组2 0只:①克氏针组:从髁间窝穿入直径1mm克氏针固定;②CPC组:将CPC从1 2号针头注入骨髓腔,手法固定至骨水泥凝固;③CPC- BMP组:将CPC与rhBMP- 2复合材料用1 2号针头注入骨髓腔,手法固定至骨水泥凝固。术后第2、4、6、8wk ,3组各取5只动物处死,行碱性磷酸酶测定,骨痂直径测量。结果 在上述4个时间点,与克氏针组和CPC组比较,CPC BMP组的碱性磷酸酶值、骨痂直径,均高于上述两组,差异具有显著性意义。结论 于骨折部位局部髓腔内注射CPC -BMP复合材料,可以促进骨质疏松大鼠骨折的愈合。 OBJECTIVE To evaluate the effects of the composite of calcium phosphate cement (CPC) and recombined human bone morphogenetic protein-2 (rhBMP-2) on the fracture healing of osteoporotic rats.METHODS 60 female osteoporotic rats were acquired 10 weeks after ovariectomy. Fractures were created in the cancellous bone of the distal right femur. All the animals were divided into 3 groups of 20 each randomly: 1) Kirschner wire group: Intramedullary Kirschner wires were made as inner fixation.2) CPC group: Injectable CPC single was injected into bone medullar through 12 # needle. The fracture parts were holded by hands until CPC harden.3) CPC-BMP group:treated as CPC group but substituted the composite of CPC/rhBMP-2 for CPC.At 2, 4, 6,8 weeks after treatment,five animals from each group were killed. The activity of Alkaline phosphatase (ALP) was quantified,the femora were harvested, and the diameter of bone callus were measured.RESULTS At the 4 time-points, the diameter of callus and the activity of ALP from CPC-BMP group were higher than those from the other two groups (P<0.05).CONCLUSION These studies indicate that injecting the composite of CPC/rhBMP-2 into the bone medullar of fracture area may improve the fracture healing of osteoporotic rats.
