超声-微波协同萃取法提取杜梨果实多糖 被引量:11

摘要 目的提取杜梨果实多糖,并测定其含量。方法采用超声-微波协同萃取法和常规水浴提取杜梨果实多糖,并用蒽酮-硫酸比色法测定多糖含量。结果超声波-微波协同萃取法比较常规水浴法提取杜梨果实多糖效果更好,两种方法提取多糖的含量分别是13.91%和12.74%;葡萄糖浓度在25.15~100.6μg·ml^(-1)范围内呈良好的线性关系,平均回收率为100.5%,RSD1.59%(n=5)。结论超声-微波协同萃取法可作为杜梨果实多糖提取的首选方法,蒽酮-硫酸比色法测定多糖含量的方法准确,重复性好。 Objective To extract the polysaccharide from the fruits of Pyrus betulaefolia Bunge, and to determine its content. Methods Ultrasonic - microwave synergistic and water bath were employed to extract the polysaccharide and anthrone - sulphuric acid colorimetry was used to determine its content. Results There existed remarkably differences between the two methods; ultra- sonic - microwave synergistic extraction was the optimal method. The polysaccharide contents of the two methods were 13.91% and 12.74% respectively. The linear range was 25.15 - 100.6 μg · ml^ -1 for glucose. The average recovery rates 100.5% (RSD 1.59% ,n =5). Conclusion The ultrasonic- microwave synergistic extraction may take the first choice method. The method of anthrone-sulphuric acid colorimetry for determination of the polysaccharide content is stable and accurate.
