摘要
研究了培养基蔗糖浓度对莲藕试管苗驯化的影响。结果表明:随培养基蔗糖浓度从3%~8%递增,试管苗含水量降低,碳水化合物含量增加,且各处理间差异明显。驯化过程中各处理试管苗淀粉含量均明显下降,低蔗糖浓度培养的试管苗可溶性总糖、蔗糖、果糖、葡萄糖含量在驯化过程中持续上升,高蔗糖浓度培养的试管苗的上述糖含量则持续下降,至成活时差异明显缩小。5%蔗糖培养的试管苗驯化过程中含水量及碳水化合物的含量变化均最小,试管苗驯化成活快,成活率最高,达97.3%,成活苗新生根数和地下茎生长速度均极显著优于其他处理,试管苗生长势旺。表明驯化前较高的培养基蔗糖浓度有利于提高试管苗的素质,提高驯化成活率。
This experiment studies the effect of medium sucrose content on the domestication of lotus root tissue culture plantlets. The results showed:carbohydrate content in plantlets was increased with distinct difference by increasing medium sucrose content from 3 % to 8 % ,while water content was decreased at the same time. Starch concentration in plantlets of different treatments was all reduced,soluble sugar,sucrose,fructose and glucose content in plantlets cultured by low sucrose concentration medium were increased, but those contents decreased in high sucrose medium. The differences reduced remarkably when they survive. Plantlets cultured in medium with 5 % sucrose had the smallest changing rate in water and carbohydrate concentration during the process of domestication, but their survival rate was the highest, up to 97.3 % , and all plantlets grew quickly and healthily. This indicates that a higher medium sucrose concentration is favorable for improving the quality and survival rate of plantlets before domestication.
