摘要
目的:建立以RP-HPLC法测定菌毒清颗粒中哈巴俄苷含量的方法。方法:色谱柱为DiamonsidTM-C18(250mm×4.6mm,5μm),流动相为乙腈-1%醋酸溶液(二元梯度洗脱),流速为1.0mL.min-1,检测波长为278nm,柱温为室温。结果:哈巴俄苷检测浓度在2.55~20.4μg.mL-1范围内与峰面积积分值呈良好线性关系;平均回收率为96.6%,RSD=0.67%(n=6)。结论:本法简便、准确,可用于菌毒清颗粒的质量控制。
OBJECTIVE: To develop a RP-HPLC method for the determination of Harpagosicle in Junduqing granule, METHODS: The chromatographic column was Diamonsil-C18(250mm× 4.6mm, 5μm) at room temperature. The mobile phase was acetonitrile-1%HAc(gradient elution) . The flow rate was 1.0mL·min^-1 and the detection wavelength was 278 nm. RESULTS: The linear range of Harpagosicle was 2,55--20,4μg ·mL^-1, with average recovery at 96.6%, RSD = 0,67% (n = 6). CONCLUSION: The method was proved to be sensitive and accurate, and it can be used as quality control of Junduqing granule.
