摘要
目的研究β-catenin及E-cadherin在肝细胞癌中的表达,探讨二者与肝癌及相关临床和病理学因素的关系。方法采用S-P免疫组化法,检测72例HCC中β-catenin及E-cadherin的表达。结果在肝硬化和癌周肝组织中β-cat和E-cad基本在细胞膜表达。肝癌组织中β-cat异常表达表现为细胞膜表达减弱、缺失和异位表达(胞浆,胞核表达),异常表达率为76.38%,与肝硬化和癌周肝组织比有明显差异(P<0.05)。肝癌组织E-cad异常表达表现为膜表达减弱或缺失,异常率为61.11%,与肝硬化和癌周肝组织比有明显差异(P<0.05)。在不同分化程度肝癌细胞中,β-cat异位表达率为:高分化63.6%,低分化96.4%,差异显著(P<0.05)。E-cad的异常表达率为:高分化47.7%,低分化85.7%,差异显著(P<0.05)。E-cad在14名有门静脉癌栓病例中表达异常率明显高于无癌栓者(P<0.05)。结论β-cat和E-cad的异常表达与肝癌有关,其中E-cadherin在肝癌细胞膜表达的缺失和减弱可能为导致β-catenin异位表达的一个因素,从而导致肝癌发生。β-cat和E-cad的异常表达与癌组织分化程度有关,E-cad的异常表达与肝癌门脉癌栓有关。β-cat和E-cad的表达与肝癌组织分型,肿瘤大小等因素无关。
Objective To investigate the expression of β - cat and E - cad in HCC and to discus the relationship between the β -cat, E- cad and liver cancer as well as related clinical and pathological factors. Methods Immunohistological method Was employed to test β - cat and E - cad expressions in 72 HCC. Results β - cat and E - cad of liver cirrhosis and paratumor express on cell membranes. Abnormal expression of β - cat in HCC includes reduction of expression in cell membrane and absent as well as abnormal located expression (expression in cytoplasm and nucleus) , abnormal rate is 76. 38% , which is significantly higher than that of liver cirrhosis and paratumor (P 〈 0.05 ) . Abnormal expression of E - cad in HCC is reduction and absence in cell membrane and the abnormal rate is 61.1%, which is significantly distant from that of liver cirrhosis and paratumor (P 〈 0. 05 ) . Abnormal located expression rate of β - cat is 63. 6% in high - differentiated HCC, 96. 4% in low - differentiated HCC, being obviously different (P 〈 0. 05) . Abnormal expression rate of E - cad is 47.7% in high - differentiated HCC, 85, 7% in low - differentiated HCC , being obviously different (P 〈 0. 05 ) . Abnormal expression rate of E - cad in 14 HCC with portal vein embolus is significantly higher than in HCC without portal embolus ( P 〈 0. 05 ) . Conclusions Abnormal expressions of β - cat and E - cad are associated with the liver cancer, the absence and reduction of E - cadherin in cell membrane of liver cancer can be a factor which leads to abnormal located expression of β - cat and causes liver cancer. Abnormal expression of E - cad and β - catare are associated with the degree of differentiation in HCC, abnormal expression of E - cad is associated with portal vein embolus. There is no relevance between expression of β - cat as well as E - cad and tumor's diameter or histological type of liver Cancer or other observed factors.
