摘要
目的:建立血浆中葛根素的高效液相色谱测定方法,为含葛根素中药复方体内代谢研究提供基础。方法:血浆用0.58mol·L-1高氯酸沉淀蛋白质,采用ShimpackCLCODS柱(5μ,150mm×4.6mm),预柱为YWGC18H37,流动相为甲醇5.625mol·L-1醋酸水(25∶3∶72),流速为1.00ml·min-1,检测波长为247nm。结果:该法回收率91.0%,方法重现性RSD=0.93%(n=6)。结论:本文建立的高效液相色谱测定方法为动物体内葛根素的测定提供了一个可靠的方法。
OBJECTIVE:To establish the determining method of puerarin in plasma with high performance liquid chromatography.METHOD:The 0.58 mol·L -1 perchloric acid was used to precipitate the protein.Shimpack CLC ODS column(5 μ,150 mm×4.6 mm) was used as analytical column.YWG C 18 H 37 was used as pre column.The mobile phase consisted of methanol 5.625 mol·L -1 acetic acid water(25∶3∶72).The rate of mobile phase was 1.00 ml.RESULTS:The recovery of this method was 91.0%,and the repeatability was RSD=0.93%(n=6).CONCLUSION:The method could be used to determine puerarin in vivo.
