摘要
目的建立测定青蒿药材及不同提取方法所得青蒿油中石竹烯和樟脑含量的方法。方法采用毛细管气相色谱法,色谱柱为HP-5石英毛细管柱(30 m×0.53 mm,1.5μm);载气为氮气;十三烷为内标物;程序升温,检测器温度270℃;N2流速3 ml.min-1,H2流速35 ml.min-1,空气流速300 m.lmin-1,尾吹气N2流速30 m.lmin-1。结果石竹烯、樟脑与内标物十三烷能达到良好分离,线性范围0.049-0.490 ng;青蒿药材中樟脑的加样回收率为97.21%(RSD=0.79%)、石竹烯为100.38%(RSD=0.73%),青蒿油中樟脑的加样回收率为98.89%(RSD=1.09%)、石竹烯为100.61%(RSD=0.87%)。结论所建方法灵敏、准确、重复性好,可用于青蒿药材及青蒿油的质量控制。
OBJECTIVE To establish a method for GC determination of camphor and caryophyllene in Artemisia annua L. and Herba Artemisiae annuae oil. METHODS Capillary GC was adopted. The internal standard method was employed with tridecane as internal standard; the GC systerm consisted of Agilent GC -6890 with a flame ionization detector, HP -5 capillary column (30 m × 0.53 mm, 1.5 μm) , nitrogen as the carrier gas ; temperature programming, detecror tempertature 270℃ , N2 : 3 ml · min ^- 1 , H2.35 ml· min ^-1, air: 300 ml·min^-1,made up( N2 ) :30 ml·min^-1. RESULTS Camphor and caryophyllene could be separated well under the chromatographic conditions. The linear range was from 0.049 ng to 0. 490 ng. The mean recovery of caryophyllene in Artemisia annua L was 97.21% with RSD of 0.79% and that of camphor was 100.38% with RSD of 0.73% , respectively. While that of Herba Artemisiae annuae oil was 98.89% with RSD of 1.09% and that of camphor was 100.61%with RSD of 0. 87% , respectively. CONCLUSION The method is sensitive, accurate, and has good repeatability, which can be used to control the quality of Artemisia annua and Herba Artemisiae annuae oil.
