多房棘球绦虫重组BCG-EmⅡ/3疫苗免疫小鼠后IgG及其亚类和IgE的动态观察 被引量:3

摘要 目的动态观察多房棘球绦虫重组BCG-EmⅡ/3疫苗免疫小鼠后IgG及其亚类和IgE的变化。方法将疫苗采用鼻腔内接种方法免疫BALB/c鼠,免疫后0、2、4、6、8、10、12、14、16和18周各剖杀4只小鼠,眼球取血,常规ELISA测定血清IgG及其亚类和IgE水平,同时设有PBS对照。结果疫苗免疫组的血清IgG、IgG2a和IgG2b水平均在免疫后2-18周升高,分别在免疫后10、4和4周达最高水平,ELISA A值分别为0.097±0.030、0.024±0.003和0.022±0.003;血清IgG1和IgE水平均在免疫后2-18周显著降低,分别在免疫后4周和18周达最低水平,A值分别为0.022±0.005和0.181±0.011;血清IgG3水平在免疫后10-12周增加,在免疫后12周达最高水平,A值为0.212±0.008。结论多房棘球绦虫重组BCG-EmⅡ/3疫苗在免疫早期即可诱导小鼠产生一个TH1型保护性免疫反应。 Objective To observe dynamically the changes of IgG, its subclass and IgE in sera of mice by immunization with recombinant BCG-EmⅡ/3 vaccine of Echinococcus multilocularis(Em). Methods BALB/c mice were intranasally vaccinated the vaccine and killed on 0, 2, 4, 6, 8, 10, 12, 14, 16 and 18 weeks of immunization respectively, sera were gathered from the eyeball to measure IgG, its subclass and IgE by routine ELISA, PBS served as control. Results In the groups of immunization, levels of IgG, IgG2a and IgG2b increased obviously on 2-18 weeks, reached the highest level on 10, 4 and 4 weeks respectively; levels of IgG1 and IgE decreased remarkably on 2-18 weeks, came to the lowest level on 4 and 18 weeks respectively; levels of IgG3 rose obviously on 10-12 weeks, achieved the peak value on 12 weeks. Conclusion TH1 response is induced in mice by rBCG-Em Ⅱ/3 vaccine on early immunization.
