摘要
【目的】设计合成肿瘤抗原MAGE-12的B细胞表位并研究其免疫原性。【方法】在预测MAGE-12的B细胞表位序列为LEQRSQHCKPEE(3~14)、MAGE-1282-93(QSDEGSSNEEQE)的基础上,采用4分枝多重抗原肽结构设计合成抗原肽,免疫C-57BL/6小鼠,产生多克隆抗体,应用ELISA实验检测多克隆抗体是否为抗原肽的特异性抗体。【结果】免疫C-57BL/6小鼠后可产生MAGE-12的抗原特异性的多克隆抗体。【结论】证明所预测的表位LEQRSQHCKPEE(3—14)为MAGE-12的新B细胞表位。
[Objective] To design and synthesis B-cell eptope of tumor antigen MAGE-12 for the study of immunogen. [Methods] Based on epitope 3 - 14 and 82 - 93, a mutiple antigenic peptide system (MAP) was synthesized as immunogen and C-57BL/6 mice were immunized with the immunogen and the polyclonal anti-serum was produced. The polyclonal antibody was identified by ELISA. [ Results] The antibody titre were reached to 1 : 256, 1 : 64 respectively, the polyclonal antibody was identified as specific antibody of the antigenic peptide. [Conclusions] The results show that on epitope 3 - 14 may be a new B cell epitope of MAGE-12.
