摘要 目的:探讨少突胶质细胞在发育过程中胞膜表达Nogo-A量的变化。方法:以A2B5﹑O4﹑O1﹑MBP为不同发育时期的少突胶质细胞的标志物,分别以抗上述标志物的抗体标记少突胶质细胞,流式细胞术分选,每组收集相同数目的细胞,再分离出每组细胞的胞膜蛋白质,Western-blot半定量检测每个细胞组Nogo-A蛋白的表达情况。结果:Western-blot检测结果显示,每组在200kDa处出现阳性蛋白条带,A2B5组蛋白量最高,其次是O4﹑O1组,MBP组蛋白含量最低。结论:在发育过程中,少突胶质细胞胞膜表面的Nogo-A蛋白的量逐渐降低。 Objective :To explore changes of Nogo-A expression in membrane of oligodendrocytes in different developmental stages.Method: A2B5.O4.O1 .MBP were used as markers of oligodendroeyte lineages,the same numbers of oligodendrocytes in each group were sorted by flow cytometry,after withdrawing membrane protein and the expression of Nogo-A in each group were analysed by Western-blot method. Result:There were positive protein straps near 200kDa in each group, and the protein content in group A2B5 was the highest, in group O4,O1 were less and in group MBP was the lowest. Conclusion:The expression of Nogo-A protein in membrane declined gradually in the development course of oligodendrocyte.
