摘要
用分光光度酶偶联速率法,测定用过氧钒烟酸络合物(POV)和二甲双胍(MET)治疗前后糖尿病鼠肝细胞质的磷酸烯醇型丙酮酸羧化酶(PEPCK)活性。结果显示:糖尿病鼠肝细胞质的PEPCK活性较正常鼠明显增加(P<0.01);POV治疗组经POV治疗后该酶活性明显受抑制(P<0.01),而MET治疗组经MET治疗后该酶活性虽有轻微降低但无统计学意义。POV和MET对正常大鼠的PEPCK均无明显影响。此结果为POV降低高血糖的分子生化机理提供理论依据,亦为探讨治疗糖尿病新的降糖药提供实验基础。
Liver phosphoenolpyruvate carboxykinase (PEPCK) activity of rat with diabetes treated by peroxovanadate nicotinic acid (POV) and metformin was determined by spectrophotometrically enzyme coupling rate method. The results show liver PEPCK activity of diabetic rat's was increased significantly, compared with that of normal rats ( P< 0.01). The rat's PEPCK activity was inhibited obviously after treated with POV for 4 weeks. The enzymatic activity of the diabetic rats treated with metformin was unobviously affected on PEPCK activity of normal rats. The results provided theoretic basis for the molecular biochemical mechanism of POV lowering hyperglycemia, and provided experimental base for exploring a new lowering hyperglycemia drug.
