洋虫成虫蛋白质化学提取方法的比较 被引量:8

摘要 洋虫成虫蛋白质提取方法及主要工艺指标筛选研究结果表明:3种方法中碱提蛋白法的蛋白质得率最高,其次为Tris-HCl提蛋白法,盐提蛋白法得率最低。碱提蛋白法的主要工艺指标为:碱液质量分数1.0%、温度80℃、时间60min、固液体积比为1∶15时蛋白质得率有最大值;盐提蛋白法:盐液质量分数1.0%、温度20℃、时间2h、固液体积比为1∶10时蛋白质得率有最大值;Tris-HCl提蛋白法:Tris-HCl液pH值8.1、抽提温度10℃、抽提时间30min、固液体积比1∶10时蛋白质得率有最大值。 The optimal method of protein extraction from adult beetles of Martianus dermestoides Chevrolat and the main technological indexes are screened out. Results show that alkaline extraction method has the highest protein extraction yield, and then followed by Tris-HCl extraction method and salt extraction method in order. The optimum extraction conditions for alkaline method with the highest protein extraction yield were obtained as alkali concentration 1.0%, temperature 80 ℃, extraction time 60 min, and the ratio of solid to liquid 1: 15. The optimal conditions for salt method were obtained as salt concentration 1.0%, temperature 20 ℃, extraction time 2 h, and the ratio of solid to liquid 1: 10. The best conditions of Tris-HCl method were obtained as pH 8.1, temperature 10 ℃, extraction time 30min, and the ratio of solid to liquid 1: 10.
