摘要
【目的】研究系统性红斑狼疮(SLE)患者骨髓基质细胞因子表达情况,及其对外周血淋巴细胞增殖反应的影响,探讨SLE的发病机制。【方法】采用ELISA方法观察SLE患者骨髓基质细胞培养液IL-6、MIP-1、IFN-γ、TGF-β等细胞因子的表达,采用四甲基偶氮唑盐比色法(MTT),观察基质细胞培养上清液对外周血淋巴细胞增殖反应的影响。【结果】SLE患者骨髓基质细胞培养液IL-6、MIP-1、IFN-γ浓度明显增高,TGF-β则降低,3种细胞因子与SLEDAI评分相关;正常人及SLE患者骨髓基质细胞上清液均对刀豆蛋白A诱导的外周血淋巴细胞增殖有明显的抑制作用,而SLE患者的上清液抑制作用明显低于正常人;在分别加入抗MIP-1、抗IFN-γ单抗后,骨髓基质细胞上清液对外周血淋巴细胞增殖的抑制作用明显增强,而加入抗TGF-β单抗后,抑制作用明显减弱。【结论】SLE患者骨髓基质细胞对外周血淋巴细胞增殖的抑制作用较弱,与其部分细胞因子异常表达有关,骨髓基质细胞可能与SLE的发病、发展有一定关系。
[Objective] To investigate expression of cytokines of bone marrow stromal cells and their effect on peripheral lymphocyte proliferation in the patients with systemic lupus erythmatosus (SLE), and explore the pathogenesis of SLE. [Methods] We detected some cytokines of SLE patients by ELISA, and determined the effect on peripheral lymphocyte proliferation of bone marrow stromal cells and cell culture supematants, and the roles of IL-6, MIP-1, IFN-γ TGF-β involved in this process by MTT assay. [Results] Expression of IL-6 in bone marrow stromal cell culture suspension of SLE was higher than that in the control group, MIP-1 and IFN-γ in the SLE patients were obviously higher than those in the control group, TGF-β was on the opposite. The later three cytokines were linear correlated to SLEDAI score. Both supematant of the normal bone-marrow stromal cell and of SLE patients showed significant suppressing effect on the peripheral lymph cell proliferation, and the suppressing effect of the SLE patients was significantly weaker than normal people. Adding anti-MIP-1 or anti-IFN-γ, the suppression got much stronger;, but anti-TGF-β was on the opposite. [ Conclusions ] Bone marrow stromal cells of SLE patients was deficient in abnormal secretion of cytokines, and its suppressing effect on proliferation of peripheral lymphocytes of the SLE patients is significantly weaker than normal people, that contributed to the pathogenesis of SLE.
