摘要
目的:建立以高效液相色谱法测定小叶黑柴胡茎叶中槲皮素、异鼠李素含量的方法。方法:色谱柱为KromasilC18(250mm×4.6mm,5μm),流动相为甲醇-0.4%磷酸溶液(55∶45),流速为1.0mL.min-1,检测波长为256nm。结果:槲皮素的线性范围为0.08~0.40μg(r=0.9996),平均回收率为101.02%(RSD=1.53%);异鼠李素的线性范围为0.06~0.30μg(r=0.9992),平均回收率为101.26%(RSD=2.95%)。结论:本法操作简便、准确、重现性好,可用于小叶黑柴胡药材的质量控制。
OBJECTIVE: To establish an HPLC method for determining the contents of Quercetin and Isorhamnetin in Bupleurum..smithii var.parvifolium Shah et Y.Li. METHODS: The HPLC was performed on Kromasil column C18 (250mm×4.6mm,5μm),using methanol- 0.4% phosphoric acid(55 : 45)as mobile phase,with flow rate at 1.0mL · min^- 1 and detection wavelength at 256nm. RESULTS: The linear range of Quercetin was 0.08-0.40μg(r = 0.999 6), with an average recovery rate of 101.02% (RSD = 1.53%); that of Isorhamnetin was 0.06-0.30μg(r = 0.999 2), with an average recovery rate of 101.26% (RSD = 2.95% ). CONCLUSION : This method is simple and accurate with good reproducibility . It is suitable for the quality control of Bupleurum.smithii var.parvifolium Shan et Y. Li.
