摘要
[目的]研究紫外线C(UVC)对热休克蛋白(heat shock proteins,HSP)27、70及90的影响。[方法]用紫外线不同照射剂量(0、10、20、40、80J/m2)来照射A549细胞株,用Western-blot检测HSP27、HSP70、HSP90的表达水平。[结果]随着UVC剂量的升高,HSP70和HSP27的表达水平先升高,达到一个高峰后又降低。HSP27表达在20J/m2时达到高峰,HSP70表达在40J/m2时达到高峰,HSP90各剂量的UVC照射下,未见显著性变化。与对照组(0J/m2)相比,在10J/m2时,HSP27和HSP70表达增加(P<0.05);在20J/m2时,HSP27表达显著增加(P<0.01),HSP70表达也继续增加(P<0.05);在40J/m2时,HSP27表达开始下降,但与对照组相比差异没有显著性(P>0.05),HSP70表达仍显著增加,与对照组相比差异有显著性(P<0.01)。在80J/m2时,HSP27表达与对照组和40J/m2组相比,差异无显著性(P>0.05),HSP70表达下降,但仍高于对照组(P<0.05)。在10、20、40和80J/m2的UVC照射下,HSP90表达变化均未见显著性差异(P>0.05)。[结论]随着UVC剂量的升高,HSP70和HSP27的表达水平先升高,达到一个高峰后又降低。HSP27,HSP70能被一定剂量范围的UVC照射所诱导,高剂量UVC照射时HSP27、HSP70的表达被抑制,但HSP90表达未发现能被诱导。
[ Objective ] To explore the expression of HSP27, HSP70, and HSP90 in A549 cells exposed to different Ultraviolet C ( UVC )irradiation. [ Methods ] A549 cells were exposed to 10, 20, 40, and 80 J/m^2 Ultraviolet C irradiation respectively, with normal cultured cells as a control. Expression of HSP27, HSP70 and HSP90 was detected by western-blot assay. [ Results ] Along with the increasing dosage of UVC irradiation, HSP27 and HSP70 levels increased to a peak then decreased. HSP27 level rose up to a peak under 20 J/m^2 of UVC irradiation but HSP70 level rose up to a peak under 40 J/m^2 of UVC irradiation. Compared with the group of 0 J/m^2, HSP70 and HSP27 levels increased significantly in the group of 10 j/m^2( P〈0.05 )and HSP27 increased very significantly in the group of 20 j/m^2( P〈0.01 ). HSP70 levels increased obviously in the group of 40 j/m^2( P〈0.01 ) but HSP27 level reduced to normal level( P〉0.05 ). In the group of 80 J/m^2, HSP70 levels decreased significantly compared with 40 J/m^2 ( P〈0.05 ) but still higher compared with the group of 0 J/m^2 ( P〈0.05 ). Significant changes of HSP90 levels were not found in this study( P〉0.05 ). [ Conclusion ] Expression levels of HSP27 and HSP70 can be induced by certain dosage of UVC irradiation. Such change in HSP90 was not found in this study.
