RECK基因与基质金属蛋白酶-2相关性在早孕滋养细胞侵袭调控中的作用 被引量:5

摘要 目的:探讨RECK基因与基质金属蛋白酶-2(matrix metalloproteinase-2,MMP-2)相关性在早孕滋养细胞浸润调控中的作用。方法:分别采用免疫组化、Westernblot法、明胶酶谱法检测52例正常妊娠(早孕组27例、足月妊娠组25例)胎盘组织中RECK表达定位、蛋白含量及MMP-2的活化比例。结果:免疫组化提示早孕、足月妊娠均有RECK蛋白表达,其主要表达在细胞滋养细胞、合体滋养细胞的胞膜和胞质。RECK表达随孕周增加而增强,早孕组RECK表达明显低于足月妊娠组(P<0.05),且在具有侵袭力的早孕锚定绒毛的细胞柱上RECK表达呈弱阳性;Westernblot检测亦显示早期妊娠RECK蛋白质含量显著低于足月妊娠(P<0.05));明胶酶谱示MMP-2的活化比例则相反,早孕组MMP-2活化比例明显高于足月妊娠组(P<0.05)。结论:MMP-2活性的表达变化与滋养细胞浸润活性呈正相关,而肿瘤抑制基因RECK的表达变化与之呈负相关,MMP-2与RECK基因相互作用在滋养细胞浸润活性调控中可能起重要作用。 Objective To study the role of the correlation of reversion-inducing-cysteine-rich protein with Kazal motifs (RECK) gene with matrix metalloproteinase-2 (MMP-2) in regulation of trophoblast invasion in early pregnancy. Methods Immunohistochemlstry, Western blot and gelatin zymography were respectively used to detect the sites of RECK expression, protein level and activated MMP-2 level in the placental tissues from 52 normal pregnant women(27 in early pregnancy and 25 in term pregnancy). Results Immunohistochemistry showed RECK was expressed in both early and term pregnancy, mainly on the cell membrane and in the cytoplasm of cytotrophoblasts and syneytiotrophoblasts. RECK expression increased with gestational age and it was significandy lower in early pregnancy than in term pregnancy (P〈0.05). RECK was poorly expressed in the cellular column of the invasive villus in early pregnancy. Western blot also showed the level of RECK protein expression in early pregnancy (mean optical density:1.35±0.14) was signiticandy lower than that in term pregnancy (mean optical density : 2.68 ± 0.26) (P 〈 0.05). In contrast, the activated MMP-2 level was decreased with the progression of gestation (P〈 0.05). The ratio of the activated MMP-2 was 0.46± 0.05 in early pregnancy and 0.10 ± 0.02 in term pregnancy. Conclusions The invasive activity of trophoblast is negatively related to the changes in MMP-2 expression but positively to the changes in the expression of the tumorinhibiting gene RECK. The interaction of RECK with MMP-2 may play an important role in regulation of the trophoblast invasion in early pregnancy.
